The Gly-Y-Z Triplet Sequence

The correct folding of these chains requires a massive repetitive sequence. Each α-chain is composed of multiple triplet sequences of Gly-Y-Z:

  • Glycine is required at every third position to allow the tight packing of the helix core.
  • Y is typically proline.
  • Z is predominantly hydroxyproline. The specific presence of hydroxyproline strongly contributes to the overall stability and rigidity of the collagen molecule.

These three distinct α-chains are then twisted around one another in a rope-like manner to produce the tightly packed triple-helical structure of collagen. The interaction of the α-chains is heavily stabilized via continuous interchain hydrogen bonding.
